Infrared absorption spectrophotometry (2.2.24).<\/p>\n<p>Comparison: brivaracetam CRS.<\/p>\n<p>B. Examine the chromatograms obtained in the test for stereoisomeric purity.<\/p>\n<p>Results: The principal peak in the chromatogram obtained with the test solution is similar in retention time to the peak due to brivaracetam in the chromatogram obtained with reference solution (a).<\/p>\n<h2>TESTS<\/h2>\n<h3>Appearance of solution<\/h3>\n<p>The solution is not more opalescent than reference suspension II (2.2.1) and not more intensely coloured than reference solution BY<sub>6<\/sub>(2.2.2, Method II).<\/p>\n<p>Dissolve 1.0 g in water R and dilute to 20.0 mL with the same solvent.<\/p>\n<h3>Stereoisomeric purity<\/h3>\n<p>Liquid chromatography (2.2.29).<\/p>\n<p>Test solution: Dissolve 25.0 mg of the substance to be examined in the mobile phase and dilute to 25.0 mL with the mobile phase.<\/p>\n<p>Reference solution (a): Dissolve 5 mg of brivaracetam CRS and 5 mg of brivaracetam impurity A CRS in the mobile phase and dilute to 5 mL with the mobile phase.<\/p>\n<p>Reference solution (b): Dissolve 2 mg of brivaracetam impurity B CRS and 2 mg of brivaracetam impurity C CRS in the mobile phase and dilute to 50 mL with the mobile phase.<\/p>\n<p>Column:<\/p>\n<p>\u2014 size: l = 0.25 m, \u00d8 = 4.6 mm;<\/p>\n<p>\u2014 stationary phase: amylose derivative of silica gel for chiral separation R (10 \u03bcm);<\/p>\n<p>\u2014 temperature: 25 \u00b0C.<\/p>\n<p>Mobile phase: anhydrous ethanol R, heptane R (20:80 V\/V).<\/p>\n<p>Flow rate: 1.0 mL\/min.<\/p>\n<p>Detection: Spectrophotometer at 205 nm.<\/p>\n<p>Injection: 20 \u03bcL.<\/p>\n<p>Run time: 1.8 times the retention time of brivaracetam.<\/p>\n<p>Identification of impurities: Use the chromatogram obtained with reference solution (a) to identify the peak due to impurity A; use the chromatogram obtained with reference solution (b) to identify the peaks due to impurities B and C.<\/p>\n<p>Relative retention: With reference to brivaracetam (retention time = about 17 min): impurity B = about 0.48; impurity C = about 0.55; impurity A = about 0.68.<\/p>\n<p>System suitability:<\/p>\n<p>\u2014 resolution: minimum 2.0 between the peaks due to impurity A and brivaracetam in the chromatogram obtained with reference solution (a); minimum 1.5 between the peaks due to impurities B and C in the chromatogram obtained with reference solution (b);<\/p>\n<p>\u2014 symmetry factor: maximum 2.0 for the peak due to brivaracetam in the chromatogram obtained with reference solution (a).<\/p>\n<p>Calculation of percentage contents:<\/p>\n<p>\u2014 for impurities A, B and C, calculate the ratio of the area of the corresponding peak to the sum of the areas of the peaks due to impurities A, B, C and brivaracetam from the chromatogram obtained with the test solution.<\/p>\n<p>Limits:<\/p>\n<p>\u2014 impurity A: maximum 2.50 per cent;<\/p>\n<p>\u2014 impurity B ((R,S)-enantiomer): maximum 0.15 per cent;<\/p>\n<p>\u2014 impurity C: maximum 0.15 per cent.<\/p>\n<h3>Related substances<\/h3>\n<p>Liquid chromatography (2.2.29).<\/p>\n<p>Test solution: Dissolve 50.0 mg of the substance to be examined in water R and dilute to 50.0 mL with the same solvent.<\/p>\n<p>Reference solution (a): Dilute 1.0 mL of the test solution to 100.0 mL with water R. Inject the solution through the septum.<\/p>\n<h4>Sulfated ash (2.4.14)<\/h4>\n<p>Maximum 0.1 per cent, determined on 1.0 g.<\/p>\n<h2>ASSAY<\/h2>\n<p>Liquid chromatography (2.2.29) as described in the test for related substances, with the following modifications.<\/p>\n<p>Injection 2 \u03bcL of the test solution and reference solutions (c), (d), (e), (f) and (g).<\/p>\n<p>System suitability:<\/p>\n<p>\u2014 the coefficient of determination (r ) calculated for the calibration curve is not less than 0.995.<\/p>\n<p>Calculate the percentage content of C<sub>11<\/sub>H<sub>20<\/sub>N<sub>2<\/sub>O<sub>2<\/sub> using the calibration curve and taking into account the assigned content of brivaracetam CRS.<\/p>\n<h2>IMPURITIES<\/h2>\n<p>Specified impurities: A, B, C.<\/p>\n<p>Other detectable impurities (the following substances would, if present at a sufficient level, be detected by one or other of the tests in the monograph. They are limited by the general acceptance criterion for other\/unspecified impurities and\/or by the general monograph Substances for pharmaceutical use (2034).
