Eur. monograph 0353)<\/em><\/p>\n<p>C<sub>21<\/sub>H<sub>28<\/sub>O<sub>5<\/sub> 360.5 50-24-8<\/p>\n<p><strong>Action and use<\/strong><\/p>\n<p>Glucocorticoid.<\/p>\n<p><strong>Preparations<\/strong><\/p>\n<p>Prednisolone Tablets<\/p>\n<p>Prednisolone Gastro-resistant Tablets<\/p>\n<h2>DEFINITION<\/h2>\n<p>11\u03b2,17,21-Trihydroxypregna-1,4-diene-3,20-dione.<\/p>\n<h3>Content<\/h3>\n<p>96.5 per cent to 102.0 per cent (dried substance).<\/p>\n<h2>CHARACTERS<\/h2>\n<h3>Appearance<\/h3>\n<p>White or almost white, crystalline, hygroscopic powder.<\/p>\n<h3>Solubility<\/h3>\n<p>Very slightly soluble in water, soluble in ethanol (96 per cent) and in methanol, sparingly soluble in acetone, slightly soluble in methylene chloride.<\/p>\n<p>It shows polymorphism (5.9).<\/p>\n<h2>IDENTIFICATION<\/h2>\n<p>First identification: A, B.<\/p>\n<p>Second identification: C, D.<\/p>\n<p>A. Infrared absorption spectrophotometry (2.2.24).<\/p>\n<p>Comparison: prednisolone CRS.<\/p>\n<p>If the spectra obtained in the solid state show differences, dissolve the substance to be examined and the reference substance separately in the minimum volume of acetone R, evaporate to dryness on a water-bath and record new spectra using the residues.<\/p>\n<p>B. Examine the chromatograms obtained in the assay.<\/p>\n<p>Results: The principal peak in the chromatogram obtained with test solution (b) is similar in retention time and size to the principal peak in the chromatogram obtained with reference solution (d).<\/p>\n<p>C. Thin-layer chromatography (2.2.27).<\/p>\n<p>Test solution: Dissolve 10 mg of the substance to be examined in the mobile phase and dilute to 10.0 mL with the mobile phase.<\/p>\n<p>Reference solution: Dissolve 10 mg of prednisolone CRS in the mobile phase and dilute to 10.0 mL with the mobile phase.<\/p>\n<p>Plate: TLC silica gel F254 plate R.<\/p>\n<p>Mobile phase: methanol R, methylene chloride R (10:90 V\/V).<\/p>\n<p>Application: 5 \u03bcL.<\/p>\n<p>Development: Over 3\/4 of the plate.<\/p>\n<p>Drying: In air.<\/p>\n<p>Detection: Spray with a solution prepared as follows: dissolve 0.25 g of 2,4-dihydroxybenzaldehyde R in glacial acetic acid R, dilute to 50 mL with the same solvent and add a mixture of 12.5 mL of sulfuric acid R and 37.5 mL of glacial acetic acid R; heat the plate at 90 \u00b0C for 35 min or until the spots appear, allow to cool and examine in daylight and in ultraviolet light at 365 nm.<\/p>\n<p>Results: The principal spot in the chromatogram obtained with the test solution is similar in position, colour and size to the principal spot in the chromatogram obtained with the reference solution.<\/p>\n<p>D. Add about 2 mg to 2 mL of sulfuric acid R and shake to dissolve. Within 5 min, an intense red colour develops. When examined in ultraviolet light at 365 nm, a brownish-red fluorescence is seen. Add this solution to 10 mL of water R and mix.<\/p>\n<p>The colour fades and there is a strong yellowish-green fluorescence in ultraviolet light at 365 nm.<\/p>\n<h2>TESTS<\/h2>\n<h3>Specific optical rotation (2.2.7)<\/h3>\n<p>+ 113 to + 119 (dried substance).<\/p>\n<p>Dissolve 0.250 g in ethanol (96 per cent) R and dilute to 25.0 mL with the same solvent.<\/p>\n<h3>Related substances<\/h3>\n<p>Liquid chromatography (2.2.29). Carry out the test protected from light.<\/p>\n<p>Solvent mixture acetonitrile R, water R (40:60 V\/V).<\/p>\n<p>Test solution (a): Dissolve 10 mg of the substance to be examined in the solvent mixture and dilute to 20.0 mL with the solvent mixture.<\/p>\n<p>Test solution (b): Dissolve 25.0 mg of the substance to be examined in the solvent mixture and dilute to 20.0 mL with the solvent mixture. Dilute 1.0 mL of the solution to 10.0 mL with the solvent mixture.<\/p>\n<p>Reference solution (a): Dissolve 5 mg of prednisolone for system suitability CRS (containing impurities A, B and C) in the solvent mixture and dilute to 10 mL with the solvent mixture.<\/p>\n<p>Reference solution (b): Dissolve 5 mg of prednisolone for peak identification CRS (containing impurities F and J) in the solvent mixture and dilute to 10 mL with the solvent mixture.<\/p>\n<p>Reference solution (c): Dilute 1.0 mL of test solution (a) to 100.0 mL with the solvent mixture. Dilute 1.0 mL of this solution to 10.0 mL with the solvent mixture.<\/p>\n<p>Reference solution (d): Dissolve 25.0 mg of prednisolone CRS in the solvent mixture and dilute to 20.0 mL with the solvent mixture.
