Melting point (2.2.14): 134 \u00b0C to 137 \u00b0C.<\/p>\n<p>B. Ultraviolet and visible absorption spectrophotometry (2.2.25).<\/p>\n<p>Test solution: Dissolve 20.0 mg in anhydrous ethanol R and dilute to 100.0 mL with the same solvent. Dilute 1.0 mL of this solution to 10.0 mL with anhydrous ethanol R.<\/p>\n<p>Spectral range: 230-350 nm.<\/p>\n<p>Absorption maximum: 252 nm.<\/p>\n<p>Shoulder about: 270 nm.<\/p>\n<p>Absorption minimum 236 nm.<\/p>\n<p>Specific absorbance at the absorption maximum 260 to 280.<\/p>\n<p>C. Dissolve the contents of a vial of fenticonazole impurity D CRS in 1 mL of this solution.<\/p>\n<p>Reference solution (e): Dissolve the contents of a vial of fenticonazole impurity E CRS in 1 mL of the mobile phase.<\/p>\n<p>Column:<\/p>\n<p>\u2014 size: l = 0.25 m, \u00d8 = 4 mm;<\/p>\n<p>\u2014 stationary phase: octadecylsilyl silica gel for chromatography R (5-10 \u03bcm).<\/p>\n<p>Mobile phase: Mix 70 volumes of acetonitrile R1 and 30 volumes of a phosphate buffer solution prepared by dissolving 3.4 g of potassium dihydrogen phosphate R in 900 mL of water for chromatography R, adjusting to pH 3.0 with phosphoric acid R and diluting to 1000 mL with water for chromatography R.<\/p>\n<p>Flow rate: 1.0 mL\/min.<\/p>\n<p>Detection: Spectrophotometer at 229 nm.<\/p>\n<p>Injection: 10 \u03bcL.<\/p>\n<p>Run time: 7.5 times the retention time of fenticonazole.<\/p>\n<p>Identification of impurities: Use the chromatogram obtained with reference solution (d) to identify the peak due to impurity D; use the chromatogram obtained with reference solution (e) to identify the peak due to impurity E.<\/p>\n<p>Relative retention: With reference to fenticonazole (retention time = about 5.7 min): impurity D = about 0.6; impurity E = about 3.7.<\/p>\n<p>Calculation of percentage contents:<\/p>\n<p>\u2014 for each impurity, use the concentration of fenticonazole nitrate in reference solution (b).<\/p>\n<p>System suitability:<\/p>\n<p>\u2014 resolution: minimum 2.0 between the peaks due to impurity D and fenticonazole in the chromatogram obtained with reference solution (d);<\/p>\n<p>\u2014 signal-to-noise ratio: minimum 5 for the principal peak in the chromatogram obtained with reference solution (c).<\/p>\n<p>Limits:<\/p>\n<p>\u2014 impurity E: maximum 0.2 per cent;<\/p>\n<p>\u2014 unspecified impurities: for each impurity, maximum 0.10 per cent;<\/p>\n<p>\u2014 total: maximum 0.3 per cent;<\/p>\n<p>\u2014 reporting threshold: maximum 0.05 per cent; disregard the peak due to the nitrate ion (retention time = about1.5 min).<\/p>\n<h3>Loss on drying (2.2.32)<\/h3>\n<p>Maximum 0.5 per cent, determined on 1.000 g by drying in vacuo at 60 \u00b0C.<\/p>\n<h3>Sulfated ash (2.4.14)<\/h3>\n<p>Maximum 0.1 per cent, determined on 1.0 g.<\/p>\n<h2>ASSAY<\/h2>\n<p>Dissolve 0.450 g in 50 mL of a mixture of equal volumes of anhydrous acetic acid R and methyl ethyl ketone R.
