Eur. 11.6 update)<\/p>\n<p>Dexamethasone Sodium Phosphate Oral Solution<\/p>\n<p>General Notices<\/p>\n<p><strong>Action and use\u00a0<\/strong><\/p>\n<p>Glucocorticoid<\/p>\n<h2>DEFINITION<\/h2>\n<p>Dexamethasone Sodium Phosphate Oral Solution contains Dexamethasone Sodium Phosphate in a suitable vehicle. The oral solution complies with the requirements stated under Oral Liquids and with the following requirements.<\/p>\n<p><strong>Content of dexamethasone sodium phosphate, C<sub>22<\/sub>H<sub>28<\/sub>FNa<sub>2<\/sub>O<sub>8<\/sub>P<\/strong><\/p>\n<p>95.0 to 105.0% of the stated amount.<\/p>\n<p>Carry out all of the following procedures protected from light.<\/p>\n<h2>IDENTIFICATION<\/h2>\n<p>A.\u2003Carry out the method for thin-layer chromatography, Appendix III A, using the following solutions. (1)\u2003To a quantity of the oral solution containing 25 mg of Dexamethasone Sodium Phosphate add sufficient methanol to produce 100 mL, centrifuge and use the supernatant liquid.<\/p>\n<p>(2)\u20030.025% w\/v of dexamethasone sodium phosphate BPCRS in methanol.<\/p>\n<h3>CHROMATOGRAPHIC CONDITIONS<\/h3>\n<p>(a)\u2003Use as the coating silica gel F254.<\/p>\n<p>(b)\u2003Use the mobile phase as described below.<\/p>\n<p>(c)\u2003Apply 5 \u00b5L of each solution.<\/p>\n<p>(d)\u2003Develop the plate to 15 cm.<\/p>\n<p>(e)\u2003After removal of the plate, dry in air and examine under ultraviolet light (254 nm).<\/p>\n<h3>MOBILE PHASE<\/h3>\n<p>20 volumes of glacial acetic acid, 20 volumes of water and 60 volumes of butanol.<\/p>\n<h3>CONFIRMATION<\/h3>\n<p>The principal spot in the chromatogram obtained with solution (1) corresponds in position and colour to that in the chromatogram obtained with solution (2).<\/p>\n<p>B.\u2003In the Assay, the principal peak in the chromatogram obtained with solution (1) has the same retention time as the peak in the chromatogram obtained with solution (2).<\/p>\n<h2>TESTS<\/h2>\n<h3>Related substances<\/h3>\n<p>Carry out the method for liquid chromatography, Appendix III D, using the following solutions in methanol (40%).<\/p>\n<p>(1)\u2003To a quantity of the oral solution containing 10 mg of Dexamethasone Sodium Phosphate add sufficient methanol (40%) to produce 100 mL and filter.<\/p>\n<p>(2)\u20030.0001% w\/v of dexamethasone sodium phosphate BPCRS.<\/p>\n<p>(3)\u20030.0001% w\/v of betamethasone sodium phosphate BPCRS.<\/p>\n<p>(4)\u20030.0025% w\/v of each of dexamethasone BPCRS and betamethasone BPCRS.<\/p>\n<p>(5)\u2003Dilute 5 volumes of solution (2) to 100 volumes.<\/p>\n<h4>CHROMATOGRAPHIC CONDITIONS<\/h4>\n<p>(a)\u2003Use a stainless steel column (10 cm \u00d7 2.0 mm) packed with octylsilyl silica gel for chromatography (3 \u00b5m) (Luna C8 is suitable).<\/p>\n<p>(b)\u2003Use gradient elution and the mobile phase described below.<\/p>\n<p>(c)\u2003Use a flow rate of 0.25 mL per minute.<\/p>\n<p>(d)\u2003Use a column temperature of 30\u00b0.<\/p>\n<p>(e)\u2003Use a detection wavelength of 240 nm.<\/p>\n<p>(f)\u2003Inject 25 \u00b5L of each solution.<\/p>\n<h4>MOBILE PHASE<\/h4>\n<p>Mobile phase A 39 volumes of methanol and 61 volumes of a 0.68% w\/v solution of potassium dihydrogen orthophosphate; adjust the pH to 3.5 with dilute orthophosphoric acid or dilute sodium hydroxide.<\/p>\n<p>Mobile phase B 41 volumes of methanol and 59 volumes of a 0.68% w\/v solution of potassium dihydrogen orthophosphate; adjust the pH to 3.5 with dilute orthophosphoric acid or dilute sodium hydroxide.<\/p>\n<table style=\"border-collapse: collapse; width: 100%;\">\n<thead>\n<tr>\n<th>Time (min)<\/th>\n<th>Mobile phase A (% v\/v)<\/th>\n<th>Mobile phase B (% v\/v)<\/th>\n<th>Comment<\/th>\n<\/tr>\n<\/thead>\n<tbody>\n<tr>\n<td>0<\/td>\n<td>100<\/td>\n<td>0<\/td>\n<td>isocratic<\/td>\n<\/tr>\n<tr>\n<td>30\u219240<\/td>\n<td>100\u21920<\/td>\n<td>0\u2192100<\/td>\n<td>linear gradient<\/td>\n<\/tr>\n<tr>\n<td>40\u219275<\/td>\n<td>0<\/td>\n<td>100<\/td>\n<td>isocratic<\/td>\n<\/tr>\n<tr>\n<td>75\u219280<\/td>\n<td>0\u2192100<\/td>\n<td>100\u21920<\/td>\n<td>linear gradient<\/td>\n<\/tr>\n<tr>\n<td>80\u219290<\/td>\n<td>100<\/td>\n<td>0<\/td>\n<td>re-equilibration<\/td>\n<\/tr>\n<\/tbody>\n<\/table>\n<h4>SYSTEM SUITABILITY<\/h4>\n<p>The test is not valid unless, in the chromatogram obtained with solution (4), the resolution factor between the peaks due to dexamethasone and betamethasone is at least 1.5.<\/p>\n<h4>LIMITS<\/h4>\n<p>In the chromatogram obtained with solution (1):<\/p>\n<p>the area of any peak due to betamethasone sodium phosphate is not greater than half the area of the principal peak in the chromatogram obtained with solution (3) (0.5%);<\/p>\n<p>the area of any secondary peak is not greater than 0.2 times the area of the principal peak in the chromatogram obtained with solution (2) (0.2%);<\/p>\n<p>the sum of the impurities is not greater than 1.0%.<\/p>\n<p>Disregard any peak due to dexamethasone and any peak with an area less than the area of the principal peak in the chromatogram obtained with solution (5) (0.05%).<\/p>\n<h3>Free dexamethasone<\/h3>\n<p>In the Assay, the area of any peak due to dexamethasone in the chromatogram obtained with solution (1) is not greater than the area of the corresponding peak in the chromatogram obtained with solution (2) (34%).<\/p>\n<h2>ASSAY<\/h2>\n<p>Carry out the method for liquid chromatography, Appendix III D, using the following solutions in methanol (45%).<\/p>\n<p>(1)\u2003To a quantity of the oral solution containing the equivalent of 8 mg of Dexamethasone Sodium Phosphate add sufficient methanol (45%) to produce 100 mL and filter through a 0.45-\u00b5m nylon filter.<\/p>\n<p>(2)\u20030.008% w\/v of dexamethasone sodium phosphate BPCRS and 0.0027% w\/v of dexamethasone BPCRS.<\/p>\n<h3>CHROMATOGRAPHIC CONDITIONS<\/h3>\n<p>(a)\u2003Use a stainless steel column (15 cm \u00d7 4.6 mm) packed with octadecylsilyl silica gel for chromatography (5 \u00b5m) (Nucleosil C18 is suitable).<\/p>\n<p>(b)\u2003Use isocratic elution and the mobile phase described below.<\/p>\n<p>(c)\u2003Use a flow rate of 1.0 mL per minute.<\/p>\n<p>(d)\u2003Use a column temperature of 30\u00b0.<\/p>\n<p>(e)\u2003Use a detection wavelength of 240 nm.<\/p>\n<p>(f)\u2003Inject 10 \u00b5L of each solution.<\/p>\n<h3>MOBILE PHASE<\/h3>\n<p>44 volumes of methanol and 56 volumes of a 0.68% w\/v solution of potassium dihydrogen orthophosphate, adjust the pH to 3.5 with dilute orthophosphoric acid or dilute sodium hydroxide.<\/p>\n<h3>SYSTEM SUITABILITY<\/h3>\n<p>The test is not valid unless, in the chromatogram obtained with solution (2), the resolution factor between the peaks due to dexamethasone sodium phosphate and dexamethasone is at least 10.0.<\/p>\n<h3>DETERMINATION OF CONTENT<\/h3>\n<p>Calculate the content of C<sub>22<\/sub>H<sub>28<\/sub>FNa<sub>2<\/sub>O<sub>8<\/sub>P using the declared content of C<sub>22<\/sub>H<sub>28<\/sub>FNa<sub>2<\/sub>O<sub>8<\/sub>P in dexamethasone sodium phosphate BPCRS and the declared content of C<sub>22<\/sub>H<sub>29<\/sub>FO<sub>5<\/sub> in dexamethasone BPCRS. Each mg of C<sub>22<\/sub>H<sub>29<\/sub>FO<sub>5<\/sub> is equivalent to 1.3158 mg of C<sub>22<\/sub>H<sub>28<\/sub>FNa<sub>2<\/sub>O<sub>8<\/sub>P.<\/p>\n<p>Calculate the total content of C<sub>22<\/sub>H<sub>28<\/sub>FNa<sub>2<\/sub>O<sub>8<\/sub>P in the oral solution from the equivalent amount of dexamethasone sodium phosphate due to Free dexamethasone and combining this with the content of dexamethasone sodium phosphate.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Edition: BP 2025 (Ph.
