It dissolves in dilute acids and in alkaline solutions.<\/p>\n<h2>IDENTIFICATION<\/h2>\n<p>First identification: A, B, D.<\/p>\n<p>Second identification: A, C.<\/p>\n<p>A. Specific optical rotation (2.2.7): + 18 to + 22 (anhydrous substance).<\/p>\n<p>Dissolve 0.25 g in a 4.2 g\/L solution of sodium hydroxide R and dilute to 25.0 mL with the same solution.<\/p>\n<p>B. Infrared absorption spectrophotometry (2.2.24).<\/p>\n<p>Comparison folic acid CRS.<\/p>\n<p>C. Thin-layer chromatography (2.2.27).<\/p>\n<p>Test solution Dissolve 50 mg of the substance to be examined in a mixture of 2 volumes of concentrated ammonia R and 9 volumes of methanol R, and dilute to 100 mL with the same mixture of solvents.<br \/>\nReference solution Dissolve 50 mg of folic acid CRS in a mixture of 2 volumes of concentrated ammonia R and 9 volumes of methanol R, and dilute to 100 mL with the same mixture of solvents.<\/p>\n<p>Plate TLC silica gel plate R.<\/p>\n<p>Mobile phase concentrated ammonia R, propanol R, ethanol (96 per cent) R (20:20:60 V\/V\/V).<\/p>\n<p>Application 2 \u03bcL.<\/p>\n<p>Development Over 3\/4 of the plate.<\/p>\n<p>Drying In air.<\/p>\n<p>Detection Examine in ultraviolet light at 365 nm.<\/p>\n<p>Results The principal spot in the chromatogram obtained with the test solution is similar in position, fluorescence and size to the principal spot in the chromatogram obtained with the reference solution.<\/p>\n<p>D. Water (see Tests).<\/p>\n<h2>TESTS<\/h2>\n<h3>Related substances<\/h3>\n<p>Liquid chromatography (2.2.29).<\/p>\n<p>Solution A 28.6 g\/L solution of sodium carbonate R.<\/p>\n<p>Test solution Dissolve 50.0 mg of the substance to be examined in 2.5 mL of solution A and dilute to 50.0 mL with the mobile phase. Dilute 2.0 mL of this solution to 10.0 mL with the mobile phase.<\/p>\n<p>Reference solution (a): Dissolve 50.0 mg of folic acid CRS in 2.5 mL of solution A and dilute to 50.0 mL with the mobile phase. Dilute 2.0 mL of this solution to 10.0 mL with the mobile phase.<\/p>\n<p>Reference solution (b): Dissolve 5 mg of folic acid for system suitability CRS (containing impurities C, E, G and H) in 1 mL of solution A and dilute to 25 mL with the mobile phase.<\/p>\n<p>Reference solution (c): Dilute 1.0 mL of the test solution to 100.0 mL with the mobile phase. Dilute 1.0 mL of this solution to 10.0 mL with the mobile phase.<\/p>\n<p>Reference solution (d): Dissolve 10.0 mg of folic acid impurity A CRS in 1 mL of solution A and dilute to 100.0 mL with the mobile phase. Dilute 1.0 mL of this solution to 100.0 mL with the mobile phase.<\/p>\n<p>Reference solution (e): Dissolve 4.0 mg of folic acid impurity D CRS in solution A and dilute to 100.0 mL with solution A.
