Eur. monograph 0179)<\/p>\n<p><strong>Action and use<\/strong><\/p>\n<p>Macrolide antibacterial.<\/p>\n<p><strong>Preparations<\/strong><\/p>\n<p>Erythromycin Gastro-resistant Capsules<\/p>\n<p>Erythromycin Gastro-resistant Tablets<\/p>\n<p>Erythromycin and Zinc Acetate Lotion<\/p>\n<h2>DEFINITION<\/h2>\n<p>Mixture of macrolide antibiotics produced by a strain of Streptomyces erythreus.<\/p>\n<p>Main component (3R,4S,5S,6R,7R,9R,11R,12R,13S,14R)-4-[(2,6-dideoxy-3-C-methyl-3-O-methyl-\u03b1-L-ribo-hexopyranosyl)oxy]-14-ethyl-7,12,13-trihydroxy-3,5,7,9,11,13-hexamethyl-6-[[3,4,6-trideoxy-3-(dimethylamino)-\u03b2-D-xylo-hexopyranosyl]oxy]-1-oxacyclotetradecane-2,10-dione (erythromycin A).<\/p>\n<p>Content<\/p>\n<p>\u2014 sum of erythromycins A, B and C: 93.0 per cent to 102.0 per cent (anhydrous substance);<\/p>\n<p>\u2014 erythromycin B: maximum 5.0 per cent (anhydrous substance);<\/p>\n<p>\u2014 erythromycin C: maximum 5.0 per cent (anhydrous substance).<\/p>\n<h2>CHARACTERS<\/h2>\n<h3>Appearance<\/h3>\n<p>White or slightly yellow powder or colourless or slightly yellow crystals, slightly hygroscopic.<\/p>\n<h2>Solubility<\/h2>\n<p>Slightly soluble in water (the solubility decreases as the temperature rises), freely soluble in ethanol (96 per cent), soluble in methanol.<\/p>\n<p>It shows polymorphism (5.9).<\/p>\n<h2>IDENTIFICATION<\/h2>\n<p>First identification: A.<\/p>\n<p>Second identification: B.<\/p>\n<p>A. Infrared absorption spectrophotometry (2.2.24).<\/p>\n<p>Comparison: erythromycin A CRS.<\/p>\n<p>Disregard any band in the region from 1980 cm to 2050 cm .<\/p>\n<p>If the spectra obtained show differences, dissolve 50 mg of the substance to be examined and of the reference substance separately in 1.0 mL of methylene chloride R, dry in vacuo at 60 \u00b0C for 3 h at a pressure not exceeding 0.7 kPa and record new spectra using the residues.<\/p>\n<p>B. Thin-layer chromatography (2.2.27).<\/p>\n<p>Test solution: Dissolve 10 mg of the substance to be examined in methanol R and dilute to 10 mL with the same solvent.<\/p>\n<p>Reference solution (a): Dissolve 10 mg of erythromycin A CRS in methanol R and dilute to 10 mL with the same solvent.<\/p>\n<p>Reference solution (b): Dissolve 20 mg of spiramycin CRS in methanol R and dilute to 10 mL with the same solvent.<\/p>\n<p>Plate: TLC silica gel plate R.<\/p>\n<p>Mobile phase: Mix 4 volumes of 2-propanol R, 8 volumes of a 150 g\/L solution of ammonium acetate R previously<br \/>\nadjusted to pH 9.6 with ammonia R, and 9 volumes of ethyl acetate R; allow to settle and use the upper layer.<\/p>\n<p>Application: 10 \u03bcL.<\/p>\n<p>Development: Over 2\/3 of the plate.<\/p>\n<p>Drying: In air.<\/p>\n<p>Detection: Spray with anisaldehyde solution R1 and heat at 110 \u00b0C for 5 min.<\/p>\n<p>Results: The principal spot in the chromatogram obtained with the test solution is similar in position, colour and size to the principal spot in the chromatogram obtained with reference solution (a), and its position and colour are different from those of the spots in the chromatogram obtained with reference solution (b).<\/p>\n<h2>TESTS<\/h2>\n<h3>Related substances<\/h3>\n<p>Liquid chromatography (2.2.29). If necessary, adjust the concentration of acetonitrile in the mobile phases and\/or the gradient to obtain the required separation.<\/p>\n<p>Calculation of percentage contents:<\/p>\n<p>\u2014 correction factors: multiply the peak areas of the following impurities by the corresponding correction factor:<br \/>\nimpurity D = 2; impurity E = 0.08; impurity F = 0.08; impurity L = 0.11;<\/p>\n<p>\u2014 for each impurity, use the concentration of erythromycin A in reference solution (c).<\/p>\n<p>Limits:<\/p>\n<p>\u2014 impurity C: maximum 3.0 per cent;<\/p>\n<p>\u2014 impurities A, B: for each impurity, maximum 2.0 per cent;<\/p>\n<p>\u2014 impurities D, E, F, H: for each impurity, maximum 1.0 per cent;<\/p>\n<p>\u2014 impurity L: maximum 0.4 per cent;<\/p>\n<p>\u2014 any other impurity: for each impurity, maximum 0.4 per cent;<\/p>\n<p>\u2014 total: maximum 7.0 per cent;<\/p>\n<p>\u2014 reporting threshold: 0.2 per cent; disregard the peaks due to erythromycins B and C.<\/p>\n<h3>Thiocyanate<\/h3>\n<p>Maximum 0.3 per cent.<\/p>\n<p>Prepare the solutions immediately before use and protect from actinic light.<\/p>\n<p>Compensation liquid: Dilute 1.0 mL of a 90 g\/L solution of ferric chloride R to 50.0 mL with methanol R.<\/p>\n<p>Test solution Dissolve 0.100 g of the substance to be examined in 20 mL of methanol R, add 1.0 mL of a 90 g\/L solution of ferric chloride R and dilute to 50.0 mL with methanol R.<\/p>\n<p>Prepare 2 independent reference solutions.<\/p>\n<p>Reference solution: Dissolve 0.100 g of potassium thiocyanate R, previously dried at 105 \u00b0C for 1 h, in methanol R and dilute to 50.0 mL with the same solvent. Dilute 5.0 mL of the solution to 50.0 mL with methanol R. To 5.0 mL of this solution, add 1.0 mL of a 90 g\/L solution of ferric chloride R and dilute to 50.0 mL with methanol R.<\/p>\n<p>Measure the absorbance (2.2.25) of each reference solution (A1, A2) and of the test solution (A) at the absorption maximum (about 492 nm).<\/p>\n<p>Suitability value:<\/p>\n<p>m<sub>1<\/sub>, m<sub>2<\/sub> = mass of potassium thiocyanate used to prepare reference solutions A1 and A2 respectively, in grams.<\/p>\n<p>The test is not valid unless S is not less than 0.985 and not greater than 1.015.<\/p>\n<p>Calculate the percentage content of thiocyanate using the following expression:<\/p>\n<p>m = mass of the substance to be examined used to prepare the test solution, in grams;<\/p>\n<p>58.08 = relative molecular mass of the thiocyanate moiety;<\/p>\n<p>97.18 = relative molecular mass of potassium thiocyanate.<\/p>\n<h4>Water (2.5.12)<\/h4>\n<p>Maximum 6.5 per cent, determined on 0.200 g.<\/p>\n<p>Use a 100 g\/L solution of imidazole R in anhydrous methanol R as the solvent.<\/p>\n<h4>Sulfated ash (2.4.14)<\/h4>\n<p>Maximum 0.2 per cent, determined on 1.0 g.<\/p>\n<h2>ASSAY<\/h2>\n<p>Liquid chromatography (2.2.29) as described in the test for related substances with the following modifications.<\/p>\n<p>Injection: Test solution and reference solutions (a) and (b).<\/p>\n<p>System suitability: Reference solution (a):<\/p>\n<p>\u2014 symmetry factor: maximum 2.5 for the peak due to erythromycin A;<\/p>\n<p>\u2014 repeatability: maximum relative standard deviation of 1.0 per cent determined on 6 injections.<\/p>\n<p>Calculate the percentage content of erythromycin A (C<sub>37<\/sub>H<sub>67<\/sub>NO<sub>13<\/sub>) using the chromatogram obtained with reference solution (a). Calculate the percentage contents of erythromycin B (C<sub>37<\/sub>H<sub>67<\/sub>NO<sub>12<\/sub>) and erythromycin C (C<sub>36<\/sub>H<sub>65<\/sub>NO<sub>13<\/sub>) using the chromatogram obtained with reference solution (b).<\/p>\n<h2>STORAGE<\/h2>\n<p>In an airtight container, protected from light.<\/p>\n<h2>IMPURITIES<\/h2>\n<p>Specified impurities A, B, C, D, E, F, H, L.<\/p>\n<p>Other detectable impurities (the following substances would, if present at a sufficient level, be detected by one or other of the tests in the monograph.
